British "Nature" magazine published two important advances in stem cell research on the 16th. Blood cells are produced by hematopoietic stem cells, which occur during embryonic development and are produced by specialized endothelial cells on the vessel wall. Hematopoietic stem cells have the long-term self-renewal ability and the potential to differentiate into various mature blood cells. It is also the longest and deepest type of adult stem cells in human history. Most leukemias are directly or indirectly related to hematopoietic stem cell abnormalities.
This time, George Dali, a researcher at Boston Children's Hospital and Dana Farber Cancer Institute, led a team that first used chemical signals to convert human pluripotent stem cells into hematopoietic endothelial cells and then changed the levels of seven key transcription factors. They are induced to become hematopoietic stem cell-like cells.
In the second paper published at the same time, Sahin Rafi and colleagues at Cornellville School of Medicine in the United States used adult mouse endothelial cells as a starting material and then changed the levels of key transcription factors to drive them to transform. In further research, both teams used environmental signals to mature hematopoietic stem cells. The Dali team transplanted human cells into the bone marrow of adult mice, and the Raf team allowed mouse cells to grow on the embryonic endothelial cell layer. The resulting cells have all the characteristics of hematopoietic stem cells - they can be transplanted into recipients to produce a variety of different blood cell lines.
